<p>I'm about to embark headfirst into Dr. K's leptin reset due to the fact that I've been steadily gaining weight and exhibit many of the LR symptoms. I have PCOS and am probably insulin resistant. My basic take on the leptin reset is that if you correct the LR the IR will follow. I'm currently taking low-dose Metformin (500 mg) and I'm wondering if this will interfere with my body resetting itself. Should I hold off or take it at a specific time?</p>
<p>A little background info- 25 y/o female, PCOS, 142 lb, was happily at 125 for a while then went on birth control to "cure" my amenorrhea, packed on 15 lbs in 2 months and have not been able to lose it. Amenorrhea returned after I quit the pill, my doctor put me on Metformin in attempt to manage PCOS. I might as well be taking a placebo, I notice no difference.</p>

<p>It leads to AMPK activation which is associated with inhibition of lipogenesis because it inactivates acetyl CoA carboxylase (ACC) and suppresses lipogenic enzyme expression. AMPK activation is also required to suppress glucose production in hepatocytes.</p>
<p>This relates closely to Dan Atkinson's theory of Adenylate energy charge</p>

I have no idea what effect Metformin will have on the Leptin Reset, but for women with PCOS, NAC outperformed Metformin and didn't have the side effects or price tag.</p>

Ive been doing it for 3 weeks now and had one day last week where I didnt stick to the protocol so I guess I am back where I started. I haven't lost any weight yet - but I haven't gained so I guess that's better than nothing....</p>
